Fig. 4. Co-localization of GFP–Wag31 and nascent peptidoglycan synthesis. Wild-type M. smegmatis cells containing Pacetgfpwag31 were cultured to exponential phase in 7H9 liquid medium with 0.1 % acetamide and stained with 1 µg Van-Alexa568 ml–1 for 3.5 h. Cells were then washed and examined by fluorescence microscopy. Images from Van-Alexa568 staining are shown in red (upper row) and GFP–Wag31 signals are shown in green (middle row). The two fluorescent images were then overlapped with the bright-field DIC image (bottom row).
